Clodronate is very effective in the treatment of hypercalcemia and cancer metastases to bones. It can significantly reduce the development of new metastases, as well as to decrease the risk of bone fractures, which is very high in such cases. Besides, it may also reduce the pain associated with this disease. Clodronate liposome, also known as clodrosome, seems to be the best way of delivering this compound to tissues.

Liposomes are lipid-based nanoparticles. Liposomal encapsulated drugs are mostly used in academic research laboratories. Some new anti-cancer compounds such as this one require a liposome delivery system to be closed in vivo. Target organs for this particular compound are kidneys and bones.

Manipulating clodronate liposome is associated with some risks, because it is highly irritating to skin, eyes, mucous membranes and respiratory system. It is very important to use appropriate clothing, chemical-resistant goggles and gloves and respirators. This suspension might be dangerous if swallowed or inhaled, and it shouldn't get in contact with your skin or eyes. It can be easily absorbed through skin.

In case of oral exposure, you should wash out mouth with water and call a physician. If your eyes were in contact with this compound, you should flush them with water for at least fifteen minutes. Dermal exposure should be treated with soap and water. If inhaled, it may cause breathing problems. Exposed person should get to fresh air, and sometimes it might be necessary to use respirator or even oxygen.

Liposomal encapsulated drugs and other compounds are required to manipulate macrophage function. Macrophages have different functions. They participate in natural immune reactions, but they also regulate the function of other, non-phagocytic cells. They ingest and digest microorganisms and macromolecules.

Liposomes encapsulate clodronate, and they are used for delivering it to tissues, in this case kidneys and bones. They will be ingested and digested by macrophages. After that, active compound will be released and accumulated. This will create macrophage depleted tissues or organs, allowing in vivo research of different functional aspects of macrophages.

Clodronate liposomes application can also be used for suppression of macrophage activity in different models of autoimmune diseases, in gene therapy and similar researches. Although this method is designed especially for in vivo research, it can also be successfully used for depletion of macrophages in vitro.

The suspension should be kept at 4 degrees of Celsius, up to one month. The important thing to remember is that it should never be frozen, or exposed to temperatures over 30 degrees. It is also important to wait until this suspension reach room temperature before injecting it to your animals. Other ways your results could be different than expected.

Always shake the suspension, other ways it won't be homogeneous, and you won't get desired results. If you use the same syringe for different animals, you won't get accurate dosages, and you might infect them. Always shave and cleanse the skin with alcohol before injecting clodronate liposome to your animal.
